    (Re)thinking Critique: Transversal and Ethico-Aesthetic Dimensions in Partaking Practices

    The article examines critique in relation to current theories pursued and addressed in contemporary art and activism. The conception of a “partaking critique” seeks to conceive critique beyond universalising abstractions and totalising gestures. By reformulating critique as a partaking practice, we set ourselves in relation to the current demands and urgencies of a world that must confront the challenges of climate change, migration flows, inequalities between the global north and south, and the mistrust of democracy. We find it important that critique results not merely in a judging and condemning analysis and the division between correct and false. A “partaking critique” deals with historical conditions and traditional formulations of critique. Critique as a partaking practice is situated, local, transversal, and reparative, and thus mobilises dispositions to act in a panorama of neo-liberal mechanisms of paralysis and paranoia. We developed our understanding of critique based on our engagement with critical examples at the crossroads of artistic and activist practices, such as Colectivo Situaciones, which in the scope of this article can only be briefly presented. We queried, shifted and transformed the concept of critique employing discourse analysis and a close reading of critical (queer)feminist concepts of the late 1980s. The perspectives taken by Donna Haraway and Eve Kosofsky Sedgwick allow us to place the focus not on negative critique but on instituent and transversal processes, and thus on rethinking the transformative potentials of ethico-aesthetic practices. The current socio-political and ecological challenges require a thinking that transverses and queers traditional valorisations of critique: we do not offer a universal, objective and strong theory, but instead favour categories of partiality, situatedness and responsibility.     PMD2HD – A web tool aligning a PubMed search results page with the local German Cancer Research Centre library collection

    BACKGROUND: Web-based searching is the accepted contemporary mode of retrieving relevant literature, and retrieving as many full text articles as possible is a typical prerequisite for research success. In most cases only a proportion of references will be directly accessible as digital reprints through displayed links. A large number of references, however, have to be verified in library catalogues and, depending on their availability, are accessible as print holdings or by interlibrary loan request. METHODS: The problem of verifying local print holdings from an initial retrieval set of citations can be solved using Z39.50, an ANSI protocol for interactively querying library information systems. Numerous systems include Z39.50 interfaces and therefore can process Z39.50 interactive requests. However, the programmed query interaction command structure is non-intuitive and inaccessible to the average biomedical researcher. For the typical user, it is necessary to implement the protocol within a tool that hides and handles Z39.50 syntax, presenting a comfortable user interface. RESULTS: PMD2HD is a web tool implementing Z39.50 to provide an appropriately functional and usable interface to integrate into the typical workflow that follows an initial PubMed literature search, providing users with an immediate asset to assist in the most tedious step in literature retrieval, checking for subscription holdings against a local online catalogue. CONCLUSION: PMD2HD can facilitate literature access considerably with respect to the time and cost of manual comparisons of search results with local catalogue holdings. The example presented in this article is related to the library system and collections of the German Cancer Research Centre. However, the PMD2HD software architecture and use of common Z39.50 protocol commands allow for transfer to a broad range of scientific libraries using Z39.50-compatible library information systems

    Do we need a Unique Scientist ID for publications in biomedicine?

    BACKGROUND: The PubMed database contains nearly 15 million references from more than 4,800 biomedical journals. In general, authors of scientific articles are addressed by their last name and forename initial. DISCUSSION: In general, names can be too common and not unique enough to be search criteria. Today, Ph.D. students, other researchers and women publish scientific work. A person may not only have one name but several names and publish under each name. A Unique Scientist ID could help to address people in peer-to-peer (P2P) networks. As a starting point, perhaps PubMed could generate and manage such a scientist ID. SUMMARY: A Unique Scientist ID would improve knowledge management in science. Unfortunately in some of the publications, and then within the online databases, only one letter abbreviates the author's forename. A common name with only one initial could retrieve pertinent citations, but include many false drops (retrieval matching searched criteria but indisputably irrelevant)

Evrim teorisi, biyolojik sistemleri doğal seleksiyon işleminin devamlı üretimi olarak tanımlamaktadır. Buna benzer olarak, Genetik Algoritmalar el ile yapıldığında uzun süren hesaplamalar yerine bilgisayar üzerinde oluşturulan evrimle mühendislik problemlerinin çözümünde etkili olmaktadır. Bu çalışmada altı açıklıklı sürekli bir kirişte maksimum açıklık ve mesnet momentlerini veren hareketli yük kombinezonlarının Genetik Algoritmalarla otomatik bir şekilde düzenlenmesi incelenmiştir. Sunulan metodun hesaplama hızı tesir çizgileri yöntemi gibi klasik yöntemlerle yapılan çözüme kıyasla oldukça yüksektir. Genetic Algorithm is a form of evolution that takes place in a computer. The selection by Genetic Algorithm operates on strings of binary digits stored in the computer's memory in much the same way that natural populations of individuals evolve. The evolution theory describes the biological systems as the continuous process of the natural selections. Similar to that, since computations made by hand are time consuming, Genetic Algorithms are effective for solving engineering problems by forming evaluation in computer. In this study, the live load arrangements for maximum bending and support moments on a continuous sixspan beam are obtained automatically by Genetic Algorithms.     Activation of PKR (double-stranded-RNA-dependent protein kinase) by DNA plasmids decreases translation, and limits the amount of recombinant protein produced by transiently transfected HEK (human embryonic kidney)-293 cells. Co-expression with Ebola virus VP35 (virus protein 35), which blocked plasmid activation of PKR, substantially increased production of recombinant TPL-2 (tumour progression locus 2)–ABIN-2 [A20-binding inhibitor of NF-κB (nuclear factor κB) 2]–NF-κB1 p105 complex. VP35 also increased expression of other co-transfected proteins, suggesting that VP35 could be employed generally to boost recombinant protein production by HEK-293 cells

    ICln ion channel splice variants in Caenorhabditis elegans: voltage dependence and interaction with an operon partner protein.

    Abstract ICln is an ion channel identified by expression cloning using a cDNA library from Madin-Darby canine kidney cells. In all organisms tested so far, only one transcript for the ICln protein could be identified. Here we show that two splice variants of the ICln ion channel can be found in Caenorhabditis elegans. Moreover, we show that these two splice variants of the ICln channel protein, which we termed IClnN1 and IClnN2, can be functionally reconstituted and tested in an artificial lipid bilayer. In these experiments, the IClnN1-induced currents showed no voltage-dependent inactivation, whereas the IClnN2-induced currents fully inactivated at positive potentials. The molecular entity responsible for the voltage-dependent inactivation of IClnN2 is a cluster of positively charged amino acids encoded by exon 2a, which is absent in IClnN1. Our experiments suggest a mechanism of channel inactivation that is similar to the "ball and chain" model proposed for the Shaker potassium channel,i.e. a cluster of positively charged amino acids hinders ion permeation through the channel by a molecular and voltage-dependent interaction at the inner vestibulum of the pore. This hypothesis is supported by the finding that synthetic peptides with the same amino acid sequence as the positive cluster can transform the IClnN1-induced current to the current observed after reconstitution of IClnN2. Furthermore, we show that the nematode ICln gene is embedded in an operon harboring two additional genes, which we termed Nx and Ny. Co-reconstitution of Nx and IClnN2 and functional analysis of the related currents revealed a functional interaction between the two proteins, as evidenced by the fact that the IClnN2-induced current in the presence of Nx was no longer voltage-sensitive. The experiments described indicate that the genome organization in nematodes allows an effective approach for the identification of functional partner proteins of ion channels

    Cutaneous Squamous Cell Carcinoma with Perineural Invasion: Report on Eight Cases and Review of the Literature

    Background: Perineural invasion (PNI) in cutaneous squamous cell carcinoma (SCC) is considered to be a negative prognostic factor. A lot of uncertainty remains regarding the classification, diagnosis, treatment and prognosis of SCC with PNI. Objective: To describe typical courses of SCC with PNI and associated findings in order to suggest an optimized diagnostic and therapeutic approach. Methods: We present eight cases of SCC with PNI, considering patient and tumor characteristics, histology, treatment and clinical course regarding local recurrence and metastasization. Results: SCC patients with PNI have a higher rate of local recurrences and greater risk for metastasization than SCC patients without PNI. Age ranged from 68 to 77 years, 6 patients were male and 2 female, with all tumors localized on the head. Three patients had chronic lymphocytic leukemia. Conclusion: Based on the data of this series and the current literature, we make suggestions for better diagnostic and therapeutic management
